I see Publisher has passwords to lock a gallery, but can we still have a master username and password like CE4?

yep. In the Publisher settings there's an option to enable a master password

I do not have anything called master password only Http Authentication and that doesn't work, I.E putting a user name in there doesn't work as a master

It should be found under Publisher > Settings : Other
I'm testing the latest beta, but I'm pretty sure this has been there all along.

HTTP Authentication is for sites using HTTP Basic Authentication. This is an additional level of protection for access to the site as a whole, or Backlight as a whole. How you would set this up is site-dependent.
